Your being quite vague there. Im assuming since your asking what
questions to ask then your client has been equally vague.
I frequently get questions like "How much for an ecommerce website?"
and i always reply with the analogy "Thats like asking a builder, 'How
much for a house?' because of course it could be a greenhouse or a
mansion.".
Basically, just ask as many (relevant) questions as you can possibly
think of, when you have lack of information thats where things get
interpreted wrongly. Cover things like what they are going to sell,
features they'd like, billing systems, how to handle shipping, VAT,
different currencies etc etc. These are the things that are relevent
specifically to me (PHP coder), but then you have questions about the
design and hosting etc.
As for charging, just try to add up how long it'll take for what they
want, multiply by your hourly rate and add 10%. Thats how i do it, but
im sure other people have different ways.
